莱芜猪肉质性状分子调控机制及雪花肉遗传改良研究

摘要: 为阐明莱芜猪肉质优势的分子调控机制并建立雪花肉高效选育技术体系,本研究以纯种莱芜猪、不同血缘杂交猪及4世代闭锁选育群体为试验材料,系统测定肌内脂肪(IMF)、48 h滴水损失、肉色、大理石纹、剪切力等核心肉质性状;结合PCR-RFLP基因分型、转录组测序与荧光定量qPCR技术,解析肉质性状关键调控基因及信号通路,并开展分子标记辅助选育(MAS)。结果表明:莱芜猪48 h滴水损失为1.44±0.14%,IMF含量达10.85±0.36%,两项指标均极显著优于大约克夏猪(P<0.01);滴水损失与IMF、肉色a*呈极显著负相关(r=-0.503、-0.570,P<0.01)。H-FABP基因HH型、A-FABP基因CD型为高IMF优势基因型,可分别使IMF含量提升0.78%、1.06%(P<0.05)。转录组分析鉴定出KLF2、IGF2、UBR1、TTBK2为滴水损失调控核心基因,ACC1、FASN、DGAT2、PLIN1为脂肪沉积关键基因。遗传参数分析显示,IMF遗传力为0.42±0.05,大理石纹遗传力为0.38±0.04,二者遗传相关达0.87±0.04。经表型选择+家系控制+分子标记综合选育4世代后,IMF由10.12±1.23%提升至13.78±1.16%,滴水损失由2.13±1.05%降至0.69±0.30%(P<0.01)。本研究明确了莱芜猪肉质性状分子调控机制,建立了可复制、可推广的雪花肉专门化品系选育技术,为我国地方猪优质化、高端化育种提供理论依据与技术支撑。

Abstract: To clarify the molecular regulation mechanism of excellent meat quality in Laiwu pigs and establish an efficient breeding system for snowflake meat, purebred Laiwu pigs, crossbred pigs with different bloodlines and 4 generation closed breeding population were used as experimental materials. Core meat quality traits, including intramuscular fat (IMF), 48 h drip loss, meat color, marbling and shear force, were systematically determined. Combined with PCR-RFLP genotyping, transcriptome sequencing and quantitative real-time PCR (qPCR), key regulatory genes and signaling pathways associated with meat quality traits were analyzed, and marker-assisted selection (MAS) was performed. The results showed that the 48 h drip loss and IMF content of Laiwu pigs were 1.44 ±0.14% and 10.85 ±0.36% , respectively, which were extremely significantly better than those of Yorkshire pigs (P<0.01). Drip loss was extremely significantly negatively correlated with IMF and meat color a* (r=-0.503,-0.570; P<0.01). The HH genotype of the H -FABP gene and the CD genotype of the A-FABP gene were dominant genotypes for high IMF, increasing IMF by 0.78% and 1.06%, respectively (P<0.05). Transcriptome analysis identified KLF2, IGF2, UBR1 and TTBK2 as core genes regulating drip loss, and ACC1, FA SN, DGAT2 and PLIN1 as key genes for fat deposition. Genetic parameter analysis indicated that the heritability of IMF and marbling was 0.42±0.05 and 0.38 ±0.04, respectively, with a genetic correlation of 0.87 ±0.04. After 4 generations of comprehensive selection integrating phenotypic selection, family control and molecular markers, IMF increased from 10.12±1.23% to 13.78±1.16%, while drip loss decreased from 2.13 ±1.05% to 0.69 ±0.30% (P<0.01). This study clarified the molecular regulation mechanism of meat quality traits in Laiwu pigs and established a reproducible breeding technology for specialized snowflake meat lines, providing theoretical and technical support for premium and high-end breeding of Chinese local pig breeds.
