不同生产阶段患子宫内膜炎母猪与健康母猪产道菌群差异分析

摘要: 子宫内膜炎通常由细菌感染引起,该病是引起母猪繁殖障碍的重要原因之一,严重影响养殖场效益。了解患有子宫内膜炎的母猪和健康母猪的产道微生物群落结构差异,将为发掘母猪子宫内膜炎的病因研究、防控以及提高母猪的繁殖能力提供基础。本研究选择19头母猪,分成3组,分别为6头健康母猪组(HS)、6头配种期患病母猪组(MP)、7头哺乳期患病母猪组(SP),收集各组母猪的阴道分泌物样品。利用纳米孔测序技术对样品中细菌16SrRNA全基因测序并评估和比较患子宫内膜炎母猪和健康母猪产道微生物群落组成与结构差异。试验数据经拼接、过滤和筛选后,共获得1359746条有效读长(Cleanreads),比对后共得到2415个可操作分类单元(OTU)。在健康母猪、繁殖初期患病母猪和哺乳期患病母猪阴道分泌物中共检测到2415种细菌,3组母猪阴道分泌物中相同的细菌有699种。与健康母猪相比,在门水平上,MP组和SP组样品中变形菌门(Proteobacteria)的相对丰度极显著增加(P<0.01),而拟杆菌门(Bacteroidetes)的相对丰度显著降低(P<0.05);在属水平上,健康母猪组与患子宫内膜炎母猪相比,患子宫内膜炎母猪组的交配期产道中不动杆菌属的相对丰度占比14.92%,哺乳期母猪组的丰度占比21.83%,健康母猪组仅占比1.22%。患病组母猪阴道菌群中,变形菌门(Proteobacteria)相对丰度极显著增加(P<0.01),可能是母猪子宫内膜炎的主要致病菌,且可能与母猪的子宫内膜炎症密切相关。本试验为母猪子宫内膜炎的后续预防和治疗提供参考。

Abstract: Endometritis is generally caused by bacterial reproductive tract infection, it is one of the important causes of reproductive disorders in sows, which seriously affected the production efficiency of breeding industry. Understanding the birth canal microbial community of endometritis sows and healthy sows are a key step in developing more effective ways to prevent the occurrence of endometritis and improve the reproductive capacity of sows. In this study, 19 samples of sow endometritis were collected and divided into 3 groups, of which 6 samples were from healthy sows, 6 samples were from mating sows and 7 samples were from lactating sows. Whole gene sequencing of nanopore 16S rRNA was used to evaluate and compare the birth canal microbiota of sows with endometritis and healthy sows. After splicing, filtering, and screening of the experimental data, a total of 1 359 746 clean reads were obtained, and 2 415 operational taxonomic units (OTUs) were identified after alignment. A total of 2 415 microbial species were detected in the vaginal secretions of healthy sows, sows with diseases during the early stage of reproduction, and sows with diseases during lactation. Among these three groups of sows, 699 bacterial species were shared in their vaginal secretions. At the phylum level, the results showed that the relative abundance of Proteobacteria in the birth canal of sows with endometritis was extremely significantly increased(P<0.01), while Bacterodietes was significantly decreased (P<0.05), compared with that of healthy sows. At the classification level, compared with the healthy sow group, the relative abundance of A cinetobacter spp. in the mating period sow group of the endometritis-affected sow group was 14.92%, while in the lactation period sow group, it was 21.83%. In the healthy sow group, it was only 1.22%. The abundance of Proteobacteria in the birth tract of sows with endometritis was extremely significantly higher than that of healthy sows(P<0.01). Proteobacteria might be the main pathogen of endometritis in sows, and might be closely related to endometrial inflammation in sows. This study provided reference for the subsequent prevention and treatment of sow endometritis.
