PRRSV防控现状及疫苗的研究进展

摘要: 猪繁殖与呼吸综合征(PRRS)是危害全球养猪业的病毒性疫病之一,每年给养猪场造成巨额经济损失。尽管减毒活疫苗(Modified live virus vaccine, MLV)应用已逾30年,但行业仍深陷只免疫而不防感染、不阻传播的困境。核心症结在于猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus, PRRSV)进化出以呼吸道黏膜为核心的多维免疫逃逸机制,而传统疫苗存在系统免疫偏向、交叉保护不足及安全风险大等3大固有缺陷,配套评价体系过度依赖血清学指标,忽视了黏膜免疫这一抵御病毒入侵的第一道防线。本文系统解析PRRSV病原学特征与黏膜免疫逃逸的分子机制,评估了传统疫苗的应用现状和局限性,并聚焦以黏膜免疫为核心的新型疫苗研发与递送技术的突破,重构“黏膜指标垣系统免疫质量垣田间传播阻断”的三维整合型评价体系。本文提出主动免疫新模式:即从被动追踪病毒变异转向主动设计黏膜靶向递送和保守抗原构建的新型疫苗策略,通过技术融合与评价体系革新,在病毒入侵阶段建立稳固的黏膜免疫屏障,以期为PRRS的有效防控乃至净化提供理论基础与可行性参考。

Abstract: Porcine reproductive and respiratory syndrome (PRRS) is one of the most viral diseases affecting the global pig industry, causing enormous annual economic losses. Although modified live virus (MLV) vaccines have been used for over three decades, the industry remains trapped in the dilemma of immunization that fails to prevent infection or block transmission. The core problem lies in the multi-dimensional immune evasion mechanisms evolved by porcine reproductive and respiratory syndrome virus (PRRSV), centered on the respiratory mucosa, while conventional vaccines suffer from three drawbacks: systemic immune bias, insufficient cross-protection, and safety risks. Moreover, the systematically evaluation systems over-rely on serological indicators, neglecting mucosal immunity—the first line of defense against viral invasion. This review systematically dissected the etiological characteristics of PRRSV and the molecular mechanisms underlying its mucosal immune evasion, evaluated the current status and limitations of conventional vaccines, and focused on breakthroughs in novel vaccine development and delivery technologies centered on mucosal immunity. It also reconstructed a three-dimensional integrated evaluation system incorporating "mucosal indicators + systemic immune quality + field transmission blockade". The core proposition of this review was to propose a new paradigm of active immune engineering: shifting from passively tracking viral variation to actively designing novel vaccine strategies based on "mucosa-targeted delivery + conserved antigen construction". By integrating technologies and innovating evaluation systems, a robust mucosal immune barrier can be established at the viral entry portal. This review provided a theoretical foundation and feasible reference for the effective control and even eradication of PRRS.
