淮南猪PDK4基因克隆、生物信息学分析及组织表达研究

摘要: 本研究旨在对淮南猪丙酮酸脱氢酶激酶4(Pyruvatedehydrogenasekinase4, PDK4)基因进行克隆与生物信息学特征分析,为后续深入开展该基因功能验证提供理论依据。以淮南猪背最长肌组织cDNA为模板,通过PCR克隆其完整CDS区序列,进行生物信息学预测分析。结果显示,淮南猪PDK4基因全长1224bp,编码407个氨基酸序列,与牛、羊、人的同源性较高。该蛋白分子量为46.17kDa,属于不稳定蛋白,包含1个支链酮酸脱氢酶激酶的Adom3功能域与1个组氨酸激酶型ATP酶C端结构域;且可能与UCP3、DKAT等蛋白具有很强相关性。qRT-PCR结果显示,PDK4基因在淮南猪各组织中存在广泛表达,其中在脂肪组织中表达较高。该研究为进一步探究PDK4基因生物学功能提供了参考。

Abstract: This study aimed to clone the Pyruvate dehydrogenase kinase 4 ( PDK4) gene from Huainan pigs and analyze its bioinformatics characteristics, thereby providing a theoretical basis for subsequent in-depth functional verification of this gene. Using cDNA from the longissimus dorsi muscle of Huainan pigs as a template, the complete CDS region of the PDK4 gene was cloned by PCR, followed by bioinformatics prediction and analysis. The results showed that the full-length CDS of the Huainan pig PDK4 gene was 1 224 bp, encoding a polypeptide of 407 amino acids, which shared high homology with cattle, sheep, and humans. The encoded protein had a molecular weight of 46.17 kDa and was classified as an unstable protein. It contained one Adom3 domain of branched-chain alpha-keto acid dehydrogenase kinase and one C-terminal domain of histidine kinase-like ATPase, and may have strong correlations with proteins such as UCP3 and DKAT. The qRT-PCR results revealed that the PDK4 gene was widely expressed in various tissues of Huainan pigs, with relatively high expression in adipose tissue. This study provided a foundation for further investigating the biological functions of the PDK4 gene.
